4-Dimethylaminopyridine, widely known as DMAP, is a highly effective nucleophilic acylation catalyst. The resonance interaction between the electron-donating dimethylamino group and the pyridine ring significantly enhances the nucleophilic activity of the ring nitrogen, greatly promoting the acylation and esterification of alcohols and amines with acids. Its catalytic activity is approximately 10⁴–10⁶ times greater than that of pyridine. Acyl transfer is a widespread reaction in nature and organic synthesis, where chiral DMAP derivatives are frequently employed as asymmetric acyl transfer catalysts.
